Very little is known about Holmes III and unknown what role he plays outside of having a grudge with the Lupin bloodline and a descendant of Sherlock Holmes. He lives in the "Flying Paradise", a house that is attached to a balloon and his personality is shown to be nervous yet also known to smile.

Fujiko Mine made Lupin III fall asleep with a sleeping pill, she removed one of his teeth and filed it down to reveal a microfilm that she showed to Holmes of a deserted island. He smiled as he believed that the island contained "Lupin's Legacy" that was hidden in the tomb of Arsène Lupin. Fujiko wanted an increase in the share of the treasure, he says that she's "fierce" yet looked worried thinking about his home.

While Fujiko knows that Lupin will come for the map, yet Holmes hope that he comes as he set out a plan. He pulled out a photo of a bra that belonged to Catherine the Great and Fujiko looked with a smile, he also explains that the bra was stolen 80 years ago and the Russian government is offering a 10 billion ruble reward that apparently the government wants to destroy the bra. Going into the drawer, he gets a second photo of an axe used by George Washington to chop down his father's cherry tree and a third photo of a pick used by King Arthur. He claims that they were also stolen 80 years ago with America and England wanting to find them.

Fujiko asks what was to do in the plan to get Lupin and Holmes explains that according to his case files from his grandfather, the person who stole it was Arsène Lupin and seriously pointed his finger directly at Fujiko. His hunch is that the items are located at the tomb and nervously paced that he made a deal with America, England and Russia so they can get the items back while himself and Fujiko gets "Lupin's Legacy". Since Lupin will turn up, the three countries had sent their operatives to kill him. This news was printed on newspapers and made Lupin annoyed who was suffering from toothache. As Lupin was talking to Goemon Ishikawa XIII, he reckoned that Holmes did it for a laugh. According to Lupin, Holmes' plan was to get him in Japan and take him to the location of the island to search for the tomb.

When Lupin got to the top of the balloon, Holmes looked up then went back to reading his book along with Fujiko. Napoleon Zero from America, James Dobon from England and Rasputan from Russia were also in the house as Goemon and Daisuke Jigen also came on board. He closed his book as the Lupin gang arrived and a paper aeroplane was sent directly towards him, Dubon spat his acidic orange seeds that caused the note to fall down directly on top of the book. Lupin jumped into the room to greet Fujiko and caused Holmes to shout at the assassins who looked out of the window, Lupin claimed that they were waiting for him to make his move. Holmes smiled and closed his eyes, then he held a paper airplane in his hand as Lupin told him to read the note. The note was an apology and mentioned that they will shave his head bald. Fujiko laughed at the thought of Holmes being bald and Lupin told him to sign the note while he wants the map as he was holding a hair clipper.

Holmes was angry knowing that the three countries are with him, he pointed his finger to Lupin claiming that the tomb and the treasures no longer belong to Lupin. Lupin however says that he doesn't want his home to crash as a shake was felt with him on the floor as Goemon using his Zantetsuken to break one of the ropes. As Holmes was ready to get up, Lupin tells him about Goemon who was to cut up his home and claims up while Holmes stayed on the floor. Holmes and Fujiko got up as they witnessed Zero lifting Goemon and putting him on the couch. Jigen managed to break Zero's sunglasses and disarm his camera causing him to be on fire. Holmes put his hands up not wanting to do anything rash as Jigen opened the door, he was surprised as Lupin and Fujiko got angry at each other then looking the other way with their eyes closed. Lupin requested for the map and the note.

As Dobon fired his acidic seeds on the floor, Holmes sat on his chair looking bored as Lupin demands to sign the note. Dobon managed to take Jigen out with a punch and melted his magnum with his acidic seeds. Holmes got up and put his hands on the desk smiling that he has the upper hand however Lupin decides to have a smoke instead, with a worry he saw Lupin take on Dobon as a needle caught his acidic seeds landing on his nose then a kick to the face to knock Dobon out. Holmes was sweating and had a nervous smile as Lupin was ready to throw him some darts, he then decided to take a feather ready to sign to note. Lupin banged his fist demanded to sign however they heard a laugh coming from another room causing Lupin to kick the door down.

Holmes looked out of the window with his binoculars and saw the island in the distance, the balloon landed on the island. As the operatives caught Lupin, jigen and Goemon, Holmes holding a torch was walking down the steps with Fujiko behind him. Zero and Dobon were unable to open the door to the tomb, Holmes got out his Colt gun and told Lupin how to open the door however Lupin was unable to as he was handcuffed. With a shake of his head, he ordered Zero to free Lupin who asked him to free his friends then he nodded his head. Lupin managed to open the door and he dashed in looking for Lupin I's treasure, he searched through the crates and was unable to find anything. Lupin tells him that his grandfather was penniless, Holmes shouted thinking that Lupin was lying but he says that he needs to give up and about the leaves planted around the tomb. Lupin tells him that Arsène Lupin tricked the natives to build him the tomb however they got angry and surrounded him.

As Lupin, Jigen and Goemon ran away, Holmes was furious putting up his fist knowing that Lupin's I treasure is somewhere and ordered the operatives to beat Lupin up but not to kill him. Holmes and Fujiko caught up and were shocked that they dealt with the operatives. Lupin tells him that his grandfather wasn't the one who stole the items but he was angry thinking that Lupin was lying as his grandfather's case files say otherwise. Knowing that he has the upper hand, Lupin says it was Sherlock Holmes disguised as Arsène Lupin who stole the items. According to Lupin II, he says that his father was angry when he heard the news. Holmes got scared thinking that what Lupin was saying wasn't true but it was and Lupin frees the operatives to go after Holmes and Fujiko with Rasputan having the hair clipper.

It turned out that Holmes and Fujiko were handcuffed on a tree branch, as the wind blew Holmes' cap away Fujiko laughed at his scalped head. At first he was shocked then got bitter by the reaction. Lupin ended up with the Flying Paradise and escaped the island.
